=== Toxicity === There are two aspects to the harmful effects of plutonium: radioactivity and heavy metal poisoning. Plutonium compounds are radioactive and accumulate in bone marrow. Contamination by plutonium oxide has resulted from nuclear disasters and radioactive incidents, including military nuclear accidents where nuclear weapons have burned. Studies of the effects of these smaller releases, as well as of the widespread radiation poisoning sickness and death following the atomic bombings of Hiroshima and Nagasaki, have provided considerable information regarding the dangers, symptoms and prognosis of radiation poisoning, which in the case of the Japanese survivors was largely unrelated to direct plutonium exposure. The decay of plutonium, releases three types of ionizing radiation: alpha (α), beta (β), and gamma (γ). Either acute or longer-term exposure carries a danger of serious health outcomes including radiation sickness, genetic damage, cancer, and death. The danger increases with the amount of exposure. α-radiation can travel only a short distance and cannot travel through the outer, dead layer of human skin. β-radiation can penetrate human skin, but cannot go all the way through the body. γ-radiation can go all the way through the body. === Prostate cancer === The backdoor pathway to DHT plays a role in the development of androgen-sensitive cancers, such as prostate cancer. In some cases, tumor cells have been found to possess higher levels of enzymes involved in this pathway, resulting in increased production of DHT. Androgen deprivation therapy (ADT) is a common treatment for prostate cancer, which involves reducing the levels of androgens, specifically T and DHT, in the body. This treatment is done through the use of medications that aim to block the production or action of these hormones. While ADT can be effective in slowing the growth of prostate cancer, it also has several drawbacks, one of which is the potential for increased production of P4 and activation of the backdoor pathway of DHT biosynthesis where P4 is a substrate. Normally, this pathway is not very active in healthy adult males, as the majority of DHT is produced through the classical pathway, which involves the direct conversion of T into DHT by one of the SRD5A isozymes. However, when T levels are reduced through ADT, the body may compensate by increasing the production of P4, which the backdoor pathway can then use as a substrate. One of the main drawbacks of this increased production of P4 leads to an increase in DHT levels, which fuel the growth of prostate cancer cells. This increased production of P4 and DHT can result in the cancer becoming resistant to ADT and continuing to grow and spread.

Navitoclax is used as mono-therapy as well as in combination with chemotherapies (paclitaxel, docetaxel, gemcitabine, and irinotecan), olaparib, erlotinib, venetoclax, and rituximab in advanced hematological malignancies (in both pediatric and adult patients) and solid tumors including ovarian cancer, breast cancer, lung cancer. Additionally, an ongoing global multi-center, randomized, open-label, phase 3 study evaluating efficacy and safety of navitoclax in combination with ruxolitinib versus best available therapy in adult patients with relapsed/refractory myelofibrosis was initiated on 31 August 2020. This activates it, giving it the ability to catalyse substrate phosphorylation.
