Chemical degradation can occur through hydrolysis, oxidation, deamidation, and aggregation, while physical changes such as precipitation or surface adsorption reduce recovery. Storage conditions are chosen to slow these processes without altering the peptide itself. Because peptides vary widely, no single condition suits every sequence, so laboratories often establish stability empirically for each batch.
Temperature is a primary factor because most degradation reactions proceed more slowly at lower temperatures. Lyophilized peptides are commonly held at -20 °C or below, although some sequences remain stable at 2–8 °C for limited periods. Moisture uptake during handling can accelerate hydrolysis, so sealed containers and desiccants are used. Solutions are generally less stable than powders and may require freezing at -80 °C or refrigeration, depending on the peptide. Repeated freeze-thaw cycles can promote aggregation even when the storage temperature is otherwise suitable.
Reconstitution is often performed with sterile water, buffer, or a water-miscible organic solvent, depending on solubility. The solvent should be added gently along the vial wall, and the solution mixed by gentle swirling rather than vigorous vortexing, which can cause foaming and surface denaturation. Some sequences require a small amount of base or acid to dissolve, followed by pH adjustment. Preparing a concentrated stock solution can simplify later dilution, but the stock itself may have limited stability. Records of solvent, concentration, and date support reproducibility.
After reconstitution, dividing the solution into single-use aliquots limits multiple warming and cooling events and reduces contamination risk. Low-binding polypropylene tubes are often preferred because peptides can adsorb to glass or untreated plastic surfaces. Filling headspace with nitrogen or argon can slow oxidation, and amber or foil-wrapped containers reduce photodegradation. Each aliquot should be labeled with peptide identity, concentration, date, and storage conditions. Frozen aliquots should be thawed quickly and kept on ice until use.

==== Tolerance ==== Tolerance and dependence are risks of chronic benzodiazepine use, and can result in doses within the therapeutic range ceasing to offer meaningful symptomatic relief after prolonged use. Tolerance develops at different rates and to different degrees to the sedative, hypnotic, anticonvulsant, muscle relaxant, and anxiolytic effects of benzodiazepines. A review of benzodiazepine tolerance concluded that it "appears that tolerance develops relatively quickly for the sedative and anticonvulsant actions of benzodiazepines, whereas tolerance to anxiolytic and amnesic effects probably does not develop at all", although the included randomized controlled trial evidence is limited to 22 weeks. A review of clonazepam in the treatment of psychiatric disorders concluded that longitudinal data supports anxiolytic benefit without tolerance during long-term use, including an open-label study finding continued benefit at three years. However, the review concludes that long-term RCT evidence is scant. A study of benzodiazepine sensitivity found that patients treated chronically with alprazolam did not differ from untreated patients in terms of anxiolytic response to diazepam, suggesting a lack of anxiolytic tolerance. However, controversy remains regarding tolerance to anxiolytic effects, with some publications reporting that there is little evidence of continued efficacy beyond 4–6 months or that dependence phenomena are common.

== Ozempic television advertisement == In 2018, pharmaceutical company Novo Nordisk began using the song in its ads for Ozempic, an injectable drug originally intended for people with type 2 diabetes, but which became popular for its off-label use as a weight-loss drug. David Paton was asked to return to Abbey Road Studios to record a new version of the song, which from a vocal standpoint is little more than his singing the opening line, replacing the words "It's magic" with "Ozempic" and otherwise adding nonverbal singing.
Euthyroid sick syndrome (ESS) is a state of adaptation or dysregulation of thyrotropic feedback control wherein the levels of T3 or T4 are abnormal, but the thyroid gland does not appear to be dysfunctional. This condition may result from allostatic responses of hypothalamus-pituitary-thyroid feedback control, dyshomeostatic disorders, drug interferences, and impaired assay characteristics in critical illness. The classical phenotype of this condition is often seen in starvation, critical illness, or patients in the intensive care unit. Similar endocrine phenotypes are observed in fetal life and in hibernating mammals. The most common hormone pattern in nonthyroidal illness syndrome is low total and free T3, elevated rT3, and normal T4 and TSH levels, although T4 and TSH suppression may occur in more severe or chronic illness. This classical pattern results from type 1 allostatic load, i.e. a stress response resulting from lacking energy, oxygen, and glutathione. An alternative phenotype with a largely inverse hormonal pattern is seen in several physiological and pathological conditions, including pregnancy, obesity, endurance training, and psychiatric diseases. It is typically associated with high-T3 syndrome, increased plasma protein binding of thyroid hormones, and an elevated set point of the homeostatic system. It represents a response to type-2 allostatic load.

Bromine has two stable isotopes, 79Br and 81Br. These are its only two natural isotopes, with 79Br making up 51% of natural bromine and 81Br making up the remaining 49%. Both have nuclear spin 3/2− and thus may be used for nuclear magnetic resonance, although 81Br is more favourable. The relatively 1:1 distribution of the two isotopes in nature is helpful in identification of bromine containing compounds using mass spectroscopy. Other bromine isotopes are all radioactive, with half-lives too short to occur in nature. Of these, the most important are 80Br (t1/2 = 17.7 min), 80mBr (t1/2 = 4.421 h), and 82Br (t1/2 = 35.28 h), which may be produced from the neutron activation of natural bromine. The most stable bromine radioisotope is 77Br (t1/2 = 57.04 h). The primary decay mode of isotopes lighter than 79Br is electron capture to isotopes of selenium; that of isotopes heavier than 81Br is beta decay to isotopes of krypton; and 80Br may decay by either mode to stable 80Se or 80Kr. Br isotopes from 87Br and heavier undergo beta decay with neutron emission and are of practical importance because they are fission products.
== History == Avasimibe was the result of a rational drug design process carried out at Parke-Davis in the early 1990s which sought to obtain orally bioavailable, water-soluble ACAT inhibitors; all such inhibitors known at the time were lipophilic and poorly absorbed when taken by mouth. This process yielded several compounds with potential, including one (designated PD 138142–15) with good solubility in water and remarkable efficacy in animal studies, but it was chemically unstable and degraded rapidly, especially in acidic environments. (Undesirable CYP450 induction was first noted at this time, in PD 138142-15 and its degradation products.) Chemical modification of PD 138142-15 and retrosynthetic analysis found that avasimibe (then codenamed CI-1011) could be easily manufactured from commercially available starting compounds, and once its efficacy was demonstrated in vitro and in rat studies, it was selected for further development. After additional safety and preclinical efficacy studies in animals, phase I clinical trials in humans began in 1997, first for hyperlipidemia (June) and subsequently for atherosclerosis (December). Phase II trials for both indications followed in 1998, and phase III trials in 2001. In October 2003, clinical development of avasimibe was discontinued. Later research discredited the concept of ACAT inhibition as a treatment for dyslipidemia and atherosclerosis, and interest in these compounds as a class waned accordingly.
The estimated prevalence is 16% in people from Cyprus, 1% in Thailand, and 3–8% in populations from Bangladesh, China, India, Malaysia and Pakistan. Estimates suggest that approximately 1.5% of the global population (80 – 90 million people) are β-thalassemia carriers. However, exact data on carrier rates in many populations are lacking, particularly in developing areas of the world known or expected to be heavily affected. Because of the prevalence of the disease in countries with little knowledge of thalassemia, access to proper treatment and diagnosis can be difficult. While there are some diagnostic and treatment facilities in developing countries, in most cases these are not provided by government services and are available only to patients who can afford them. In general, poorer populations only have access to limited diagnostic facilities and blood transfusions. John Barry Larson (born July 22, 1948) is an American politician and businessman who has been serving as the U.S. representative for Connecticut's 1st congressional district since 1999. The district is based in the state capital, Hartford. A member of the Democratic Party, Larson chaired the House Democratic Caucus during the 111th and 112th United States Congress. Larson was first elected to Congress in 1998, serving fourteen consecutive terms in the House. He attempted to seek a fifteenth term, but he was defeated in the 2026 Democratic primary by former Hartford mayor Luke Bronin.
