Solutions are generally less stable than powders and may require freezing at -80 °C or refrigeration, depending on the peptide. Repeated freeze-thaw cycles can promote aggregation even when the storage temperature is otherwise suitable.
Light, oxygen, and pH influence peptide integrity through specific side-chain reactions. Methionine and cysteine residues are susceptible to oxidation, and tryptophan can degrade under strong light. Inert gas overlays and amber glass or opaque containers reduce these risks. pH affects charge, solubility, and the rate of deamidation or aggregation; a value that minimizes one pathway may increase another. The optimal pH and buffer for a given peptide are often determined experimentally, and open questions remain about predicting stability from sequence alone.

The cartilaginous pyramid of the nose ==== The cartilaginous septum (septum nasi) extends from the nasal bones in the midline (above) to the bony septum in the midline (posteriorly), then down along the bony floor. The septum is quadrangular; the upper half is flanked by two triangular-to-trapezoidal cartilages: the upper lateral-cartilages, which are fused to the dorsal septum in the midline, and laterally attached, with loose ligaments, to the bony margin of the pyriform (pear-shaped) aperture, while the inferior ends of the upper lateral-cartilages are free (unattached). The internal area (angle), formed by the septum and upper lateral-cartilage, constitutes the internal valve of the nose; the sesamoid cartilages are adjacent to the upper lateral-cartilages in the fibroareolar connective tissue. Beneath the upper lateral-cartilages lay the lower lateral-cartilages; the paired lower lateral-cartilages swing outwards, from medial attachments, to the caudal septum in the midline (the medial crura) to an intermediate crus (shank) area. Finally, the lower lateral-cartilages flare outwards, above and to the side (superolaterally), as the lateral crura; these cartilages are mobile, unlike the upper lateral cartilages. Furthermore, some persons present anatomical evidence of nasal scrolling—i.e., an outward curving of the lower borders of the upper lateral-cartilages, and an inward curving of the cephalic borders of the alar cartilages.
==== FAD ==== Flavin adenine dinucleotide in its oxidized form, FAD is a cofactor of flavoprotein oxidoreductase enzymes. Flavoprotein pyridine nucleotide cytochrome reductases, including FAD catalyse the interchange of reducing equivalents (H+ or electrons). Initial electron donors and final electron acceptors comprise single electron carriers and two electron carrying nicotinamide dinucleotides respectively.
E. coli is one of the most widely used expression hosts, and DNA is normally introduced in a plasmid expression vector. The techniques for overexpression in E. coli are well developed and work by increasing the number of copies of the gene or increasing the binding strength of the promoter region so assisting transcription. For example, a DNA sequence for a protein of interest could be cloned or subcloned into a high copy-number plasmid containing the lac (often LacUV5) promoter, which is then transformed into the bacterium E. coli. Addition of IPTG (a lactose analog) activates the lac promoter and causes the bacteria to express the protein of interest. E. coli strain BL21 and BL21(DE3) are two strains commonly used for protein production. As members of the B lineage, they lack lon and OmpT proteases, protecting the produced proteins from degradation. The DE3 prophage found in BL21(DE3) provides T7 RNA polymerase (driven by the LacUV5 promoter), allowing for vectors with the T7 promoter to be used instead.

=== Everton bid === In September 2023, 777 Partners agreed to buy Farhad Moshiri's 94.1% shareholding in English Premier League team Everton F.C. In April 2024, with the deal not complete and after the 777-owned Australian airline Bonza entered voluntary administration, Everton called upon the services of insolvency advisers Teneo. Amid lawsuits against 777 Partners in other countries, the Everton Shareholders' Association wrote to Moshiri requesting that he terminate the deal. 777 enlisted B. Riley Financial for advice to complete the deal. According to 777's own lawyers, Wander and Pasko resigned as managers of the company on May 6, 2024, but remained as its 100% owners. On June 1, 2024, the deadline for the deal to be completed expired.
